“A homemade svFCS setup installed in Poland!

Tomasz Trombik, a former member of the Hai Tao HE & Didier MARGUET's team at the Centre d'Immunologie de Marseille-Luminy, has obtained an assistant professorship at the Faculty of Biotechnology, University of Wroclaw to develop a new project on membrane proteins. Tomasz asked us to set up a spot variation Fluorescence Correlation Spectroscopy (svFCS) like the one historically developed by H&M lab and the MOSAIC team from Institut Fresnel.

In March 2018, Sébastien Mailfert and Sophie Brustlein, a former member of the Hai Tao HE & Didier MARGUET's team at the CIML, now working for CENTURI, have successfully installed an operational svFCS microscope. In the absence of an optical physics lab nearby or a possibility to have spare parts in case of missing parts (and that happened!), the implementation of this system in ten days has been a real challenge.

Spot variation Fluorescence Correlation Spectroscopy (svFCS) is a very sensitive technique based on the analysis on the fluorescence fluctuations recorded in a confocal volume with high temporal (μs) and spatial (~200nm) resolutions. This allows the determination of the diffusion characteristics of fluorescently labeled molecules and their concentration. Pioneer in this technique, the H&M team has explored the role of the membrane organization in the control of different receptors of the immune cells.”
